Idunno if this is still relevant after 4 years, but today I encountered the same problem, and after much frustration it turned out to be solved really easily for me: all I needed to do was go to C:\Program Files (x86)\Ubisoft\Assassin's Creed II and then right click and open the game as an administrator.
